Raf-1 (c-Raf)는 MAP kinase kinase kinase (MAP3K)로서 Ras 계열 GTPase 하위에서 작용하며, MEK1 및 MEK2를 인산화하여 활성화시킵니다. MEK1/2는 다시 ERK1 및 ERK2를 활성화하여 세포 분열, 세포사, 분화, 이동 등 다양한 생리적 과정에 관여합니다. Raf-1 유전자의 돌연변이는 Noonan syndrome 5와 LEOPARD syndrome 2와 관련이 있습니다.
