Target Information
CD31 (platelet endothelial cell adhesion molecule-1, PECAM-1) is an inhibitory coreceptor involved in the regulation of T cell and B cell signaling through dual immunoreceptor tyrosine-based inhibitory motifs (ITIMs). Upon phosphorylation by associated kinases, these motifs provide docking sites for protein-tyrosine phosphatases. CD31 is widely expressed within the vascular compartment, primarily at junctions between adjacent endothelial cells.
The N-terminal Ig-like domain mediates homophilic binding, crucial for cell–cell interactions. CD31 plays diverse roles in:
- Integrin-mediated cell adhesion
- Transendothelial migration
- Angiogenesis
- Apoptosis
- Negative regulation of immunoreceptor signaling
- Autoimmunity and macrophage phagocytosis
- IgE-mediated anaphylaxis and thrombosis
