Target Information
Apoptosis is related to many diseases and developmental processes. Caspase-9 plays a central role in cell death induced by various apoptosis activators. When cytochrome c is released from mitochondria, it binds to Apaf-1 to form an apoptosome, which activates procaspase-9. Activated caspase-9 subsequently activates effector caspases (caspase-3, -6, and -7), leading to proteolytic cleavage of key proteins during apoptosis.
PHAP proteins, including PHAP III, have been identified as important regulators of mitochondrial apoptosis. PHAP facilitates apoptosome-mediated caspase-9 activation and stimulates the mitochondrial apoptotic pathway. It also opposes Ras- and Myc-mediated cell transformation.
