Target Information
FAS (Fas, CD95, APO-1)는 TNFR (tumor necrosis factor receptor) 계열에 속하는 46 kDa 막관통 당단백질로, 세포사멸 수용체로 기능합니다. 약 26 kDa의 용해형 형태로도 존재합니다. FAS 자극은 FADD(FAS-associated death domain)의 응집, DISC(death-inducing signaling complex) 형성, 그리고 caspase 활성화를 유도합니다. 이 복합체의 caspase 자동 절단 과정은 세포사멸(apoptosis)로 이어지는 하위 신호 전달을 촉발합니다. FAS는 NF-kappaB, MAPK3/ERK1, MAPK8/JNK를 활성화하며, 정상 섬유아세포 및 T세포에서 증식 신호 전달에도 관여합니다. 최소 8개의 스플라이싱 변이체가 알려져 있으며, 막관통 영역이 결여된 isoform은 전체 길이 단백질이 매개하는 세포사멸을 억제할 수 있습니다. FAS/FAS ligand 시스템은 AIDS, 간염, 암 등 다양한 질환과 관련이 있으며, 항바이러스 면역반응에서도 중요한 역할을 하는 것으로 알려져 있습니다.
