Target Information

Progression through the cell cycle requires activation of cyclin-dependent kinases (Cdks). The monomeric catalytic subunit Cdk2, essential for cell cycle initiation, is inactive until bound by regulatory cyclins such as cyclin D1 and phosphorylated at Thr 160. The enzyme responsible for this phosphorylation, Cdk-activating kinase (CAK), includes a catalytic subunit, a regulatory subunit, and a subunit of unknown function. The regulatory subunit, cyclin H, is required for activation of Cdk7. Mat1, a protein associated with the cyclin H–Cdk7 nuclear complex, is essential for forming an active Cdk–cyclin complex and displays kinase activity toward both Cdk activation domains and the C-terminal domain of RNA polymerase II.
