Target Information
ICAD (Inhibitor of Caspase-3-Activated DNase) is a substrate that regulates nuclear apoptosis. It has two isoforms:
- ICAD-L/DFF45 (45 kDa): functional isoform
- ICAD-S/DFF35 (35 kDa): non-functional isoform
ICAD binds and inhibits CAD (Caspase-Activated DNase). Upon caspase-3 activation, ICAD is cleaved at residues 117 and 224, releasing CAD to perform apoptotic DNA fragmentation. ICAD also functions as a CAD-specific folding chaperone.
Recent studies suggest ICAD as a potential target for restoring normal apoptotic signaling in colon and brain cancer cells. Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
