Product Specific Information

Description:
The MEM-15 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, macrophages, and some dendritic cells. Complexes of LPS and LBP bind with high affinity to monocytes via CD14.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:

• Flow cytometric analysis.

Applications Tested:

• Pre-diluted and tested for flow cytometric analysis of normal human peripheral blood cells.
• Recommended use: 5 µL (1.0 µg)/test (100 µL final volume).
• Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton technology with engineered nucleic acid dye structures designed to minimize spillover and spread.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide receptor and exists in both membrane-bound and soluble forms. Soluble CD14 (sCD14) discriminates structural differences between LPS molecules and contributes to immune modulation. It also binds apoptotic cells and is implicated in allergic, inflammatory, and infectious processes.
Diseases associated with CD14 dysfunction include Mycobacterium chelonae infection and croup.
