Target Information
Neuron specific enolase (NSE, ENO1, ENO2, ENO3) is an enzyme that catalyzes the conversion of 2-phosphoglycerate to phosphoenolpyruvate in the glycolytic pathway, and the reverse reaction in gluconeogenesis. NSE is highly stable in biological fluids and diffuses easily into extracellular medium and cerebrospinal fluid (CSF) upon neuronal membrane injury.
NSE exists as three mammalian enolases — ENO1 (alpha), ENO2 (gamma), and ENO3 (beta).
- Alpha-subunit: expressed in most tissues
- Beta-subunit: expressed only in muscle
- Gamma-subunit: expressed primarily in neurons and neuroendocrine cells
Co-expression of NSE and chromogranin A is common in neuroendocrine neoplasms.
Antibodies to NSE protein are useful for identifying neuronal cell bodies, developing neuronal lineage, and neuroendocrine cells. Release of NSE from damaged neurons into CSF and blood is used as a biomarker of neuronal injury.
Clinically, NSE serves as a sensitive marker of neuronal damage in disorders such as stroke, hypoxic brain damage, status epilepticus, Creutzfeldt-Jakob disease, and herpetic encephalitis. Elevated NSE levels are also found in plasma and neoplasias including pediatric neuroblastoma and small cell lung cancer.
