Target Information

Serine-threonine kinase that transduces inflammatory and cell-death signals (programmed necrosis) following death receptor ligation, activation of pathogen recognition receptors (PRRs), and DNA damage.
Upon TNFR1 activation by TNF-alpha family cytokines, TRADD and TRAF2 are recruited to the receptor.
Phosphorylates DAB2IP at Ser-728 in a TNF-alpha-dependent manner, activating the MAP3K5-JNK apoptotic cascade.
Ubiquitination by TRAF2 via Lys-63-linked chains enhances downstream signaling to MAPK and NF-kappa-B pathways, mediating inflammatory gene activation.
Polyubiquitinated RIP1 binds IKBKG/NEMO to activate NF-kappa-B.
Interaction with RHIM-containing adapters triggers gene activation and necrosis. RIPK1–RIPK3 association forms a necrosis-inducing complex.
