Target Information
STAT3는 STAT (signal transducers and activators of transcription) 단백질 계열에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동 후 전사인자로 작용합니다. CREB-binding protein 등 공활성화 인자가 전사 활성화에 필요하며, Interferon-α, Interferon-γ, EGF, PDGF, IL-6 등에 의해 활성화됩니다. Tyr705 잔기에서의 인산화는 STAT3 이합체 형성, 핵 이동 및 DNA 결합에 필수적입니다. 인간 STAT3 유전자는 17번 염색체 q arm에 위치하며, 세포 성장 및 apoptosis 등 다양한 세포 과정에 관여합니다. Rac1은 STAT3 활성 조절에 관여하며, PIAS3는 STAT3의 특이적 억제 단백질입니다. 세 가지 대체 스플라이싱 변이체가 보고되었으며, STAT3 유전자 결손 생쥐는 배아 단계에서 치명적입니다.
