Target Information
Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of plasma proteins. It inhibits amyloid fibril formation by several aggregation-prone proteins (APP, APOC2, B2M, CALCA, CSN3, SNCA, LYZ variants) in vitro, without requiring ATP. It maintains partially unfolded proteins for refolding by chaperones such as HSPA8/HSC70. The secreted isoform protects cells from apoptosis and complement-mediated cytolysis. Intracellular isoforms interact with ubiquitin and SCF E3 ligase complexes to promote proteasomal degradation of target proteins, modulate NF-kappa-B activity, and regulate apoptosis and cell proliferation.
