Target Information

JMY acts both as a nuclear p53/TP53 cofactor and a cytoplasmic regulator of actin dynamics depending on conditions.
In the nucleus, it increases p53/TP53 response via interaction with p300/EP300, enhancing transcription and apoptosis during stress responses such as DNA damage.
In the cytoplasm, JMY serves as a nucleation-promoting factor for both branched and unbranched actin filaments. It activates the Arp2/3 complex to induce branched actin networks and can also catalyze actin polymerization independently of Arp2/3, promoting unbranched filaments.
This dual activity contributes to cell motility and actin dynamics regulation. The cofactor and actin activator functions are controlled by subcellular localization.
