Specificity Comments:
CD162 glycoprotein acts as a high-affinity counter-receptor for P-, E-, and L-selectin expressed on myeloid cells and activated T lymphocytes. It plays a key role in leukocyte trafficking during inflammation by mediating tethering of leukocytes to activated platelets or endothelia expressing selectins. High-affinity binding requires tyrosine sulfation and the addition of the sialyl Lewis x tetrasaccharide (sLex) to its O-linked glycans. Aberrant expression or polymorphisms in this gene are linked to innate and adaptive immune response defects.
Target Information
CD162 (PSGL-1) is a 120 kDa homodimer protein located on leukocytes and platelets. It binds to CD62P (P-selectin), CD62L (L-selectin), and CD62E (E-selectin) with varying affinities. These interactions are crucial for leukocyte adhesion, rolling, and extravasation. CD162 also serves as an immune marker of T cell exhaustion in chronic viral infections and cancer.
