Target Information
Guanylate cyclases belong to the adenylyl cyclase class-4/guanylyl cyclase family.
There are two forms of guanylate cyclase:
- Soluble form (GCS or sGC) – acts as a receptor for nitric oxide
- Membrane-bound receptor form (GC) – acts as a peptide hormone receptor
The GC-C protein consists of:
- Extracellular domain
- Single transmembrane domain
- Cytoplasmic region (kinase-like + catalytic domains)
It is expressed as two glycosylated forms:
- 130 kDa precursor (endoplasmic reticulum)
- 145 kDa mature form (plasma membrane)
Ligand binding to the extracellular domain promotes cGMP accumulation.
GC-C acts as the receptor for heat-stable enterotoxins from pathogenic E. coli, causing secretory diarrhea, and also binds guanylin and uroguanylin peptides that modulate renal function in response to salt load.
