배경 정보

cf-DNA는 암 조기 진단 및 질병 모니터링에 유용한 바이오마커 정보를 포함합니다. 소변은 비침습적이며 반복 채취가 가능하다는 장점이 있습니다. 또한 단백질 농도가 낮아 정제가 용이하고, 혈장이나 혈청과 유사한 핵산 프로파일을 가집니다. 이러한 특성으로 소변 cf-DNA는 암, 자가면역질환, 태아 DNA 연구 등에 활용되고 있습니다.

2–10 mL의 소변을 두 단계 컬럼 공정으로 처리하여 50 µL로 농축

1단계

Cell-Free Urine
Add Binding Solution K & vortex
Transfer to column → Wash → Discard flowthrough
Elute in 0.4 mL
Add Proteinase K and incubate at 55°C for 10 min

2단계

Add Lysis Buffer A, Ethanol & vortex
Transfer to mini spin column
Spin & repeat
Wash 2 times
Elute 50 µL (optionally recycle elution for maximum recovery)
DNA ready for downstream analysis

10 mL, 20 mL, 30 mL 소변에서 순환 DNA를 정제 후 qPCR 분석을 수행하였으며, 5S rRNA 유전자 양은 시료량 증가에 따라 선형적으로 증가하였습니다.

모든 용액은 밀봉 상태로 실온 보관하며, 출하일로부터 2년간 안정합니다. Proteinase K는 완충 저장 용액에 용해되어 있으며, 실온에서 2년간 안정합니다.
