Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, playing crucial roles in regulating cell death and stress responses. TRAF proteins share a homology region that enables binding to cell receptors and other TRAF proteins, activating distinct signaling cascades. TRAF2 and TRAF3 bind to CD40, a member of the NF receptor superfamily involved in B cell immunity, and are essential for NF-kappa-B activation in mouse B lymphocytes. TRAF2 and TRAF6 are required for CD40 signaling in non-hematopoietic cells. Additionally, TRAF2 interacts with the lymphotoxin-beta receptor (LTbetaR) along with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
