Target Information
MERTK (cMER) is a tyrosine kinase proto-oncogene involved in the retinal pigment epithelium (RPE) phagocytosis pathway, which is implicated in human retinal disease.
It regulates physiological processes including cell survival, migration, differentiation, and efferocytosis. Ligand binding induces autophosphorylation of MERTK and provides docking sites for downstream signaling molecules such as GRB2 or PLCG2, leading to phosphorylation of MAPK1, MAPK2, FAK/PTK2, or RAC1.
MERTK signaling contributes to macrophage clearance of apoptotic cells, platelet aggregation, cytoskeleton reorganization, and phagocytosis in RPE. It also inhibits Toll-like receptor (TLR)-mediated innate immune responses by activating STAT1, which induces SOCS1 and SOCS3 expression.
