Target Information
MERTK (also known as cMER) is a tyrosine kinase proto-oncogene involved in retinal pigment epithelium (RPE) phagocytosis, which is linked to human retinal disease. It regulates cell survival, migration, differentiation, and efferocytosis (phagocytosis of apoptotic cells). Ligand binding induces autophosphorylation of MERTK, creating docking sites for downstream signaling molecules such as GRB2 and PLCG2, leading to activation of MAPK1, MAPK2, FAK/PTK2, and RAC1 pathways. MERTK signaling is essential for macrophage clearance of apoptotic cells, platelet aggregation, cytoskeleton reorganization, and suppression of Toll-like receptor (TLR)-mediated immune responses via STAT1 activation, which upregulates SOCS1 and SOCS3 expression.
