Target Information
The inositol hexakisphosphate kinase family members, IP6K1 and IP6K2, have high affinity and selectivity for inositol hexakisphosphate (InsP6) as a substrate. They convert InsP6 to PP-InsP5 but show no catalytic activity with other inositol pyrophosphates. InsP6 inhibits serine/threonine protein phosphatases and increases calcium influx across the plasma membrane, suggesting a role in insulin exocytosis regulation. IP6K1 was purified as a 54 kDa protein in rat brain extracts, while IP6K1 and IP6K2 were characterized in mouse as 50 kDa and 49 kDa proteins, respectively. IP6K1 also displays ATP synthase activity, transferring a phosphate from PP-InsP5 to ADP, indicating a potential role as a high-energy phosphate donor.
