Product Specific Information

For reconstitution, add 100 µL distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. Upon activation, p53 upregulates genes such as p21waf1, leading to G1 or G2/M phase arrest.

The p53 structure includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation.
p53 plays a key role in preventing proliferation of damaged cells and acts as an anticancer agent. It is expressed at low levels in normal cells but upregulated in transformed cell lines. Mutations in TP53 lead to loss of tumor suppressor function and are common in various cancers, including hereditary Li-Fraumeni syndrome.
p53 is also implicated in aging, oxidative stress, and cognitive function through its influence on long-term potentiation (LTP).
