Target Information

Hypermethylated in cancer (HIC-1) was originally identified as a target of p53-induced gene expression.
HIC-1 is deleted in the genetic disorder Miller-Dieker syndrome (MDS), and its expression is frequently suppressed in leukemia and various cancers due to hypermethylation of specific DNA regions and transcriptional silencing.
Studies indicate that HIC-1 acts as a putative tumor suppressor protein mediating transcriptional repression.
HIC-1 is ubiquitously expressed in adult tissues and contains five zinc fingers and an N-terminal POZ/BTB domain.
This domain interacts with transcriptional repression complexes (SMRT/N-CoR-mSin3A HDAC), similar to BCL-6 and PLZF proteins.
However, HIC-1 does not directly interact with HDAC complex components, suggesting its repression mechanism is independent of the POZ/BTB domain.
