Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. Upon activation, p53 induces transcription of target genes such as p21waf1, leading to G1 or G2/M arrest.

Structurally, p53 contains:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization. p53 functions as a key regulator preventing proliferation of damaged cells and acts as an anticancer agent. Alterations or mutations in the TP53 gene are common in human cancers and can lead to loss of tumor suppressor activity, contributing to malignancy. Additionally, p53 is implicated in aging, oxidative stress, and neuronal function, including long-term potentiation (LTP).
