Target Information

Inositol 1,4,5-trisphosphate (IP3) acts as a second messenger for numerous growth factors, hormones, and neurotransmitters. Upon binding to the IP3 receptor (IP3R), IP3 induces the release of luminal calcium into the cytosol. The IP3R functions as a homo- or heterotetramer of ~240 kDa glycoprotein subunits and is structurally and functionally related to the ryanodine receptor. Both receptors can associate with the immunophilin protein FKBP12, influencing calcium release through phosphorylation and dephosphorylation mechanisms involving PKC and calcineurin. Mammalian IP3R subunits are encoded by three distinct genes with differential expression patterns:

  • IP3R-I: heart, liver, kidney, ovary, cerebellar Purkinje neurons
  • IP3R-II: brain
  • IP3R-III: pancreatic islets, kidney, gastrointestinal tract
