Target Information

Tau is a neuronal microtubule-associated protein predominantly found on axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules, while the N-terminus binds neural plasma membrane components, acting as a linker between both. Axonal polarity is influenced by Tau/MAPT localization within neuronal cells.

Hyper-phosphorylated Tau is the major component of paired helical filaments (PHF), which form neurofibrillary lesions in Alzheimer’s disease brains. This modification impairs Tau’s microtubule-binding ability, leading to microtubule destabilization and neuronal degeneration. Tau phosphorylation is mediated by several kinases, including GSK-3β, PKA, cdk5, and casein kinase II. Abnormal Tau phosphorylation is also associated with other neurodegenerative disorders such as Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
