Target Information

TMED2 is a protein coding gene involved in vesicular protein trafficking. It mainly functions in the early secretory pathway and post-Golgi membranes. TMED2 acts as a cargo receptor for secretory molecules and participates in vesicle coat formation.

In COPII vesicle-mediated anterograde transport, it assists in the transport of GPI-anchored proteins, working with TMED10 as a cargo receptor involving SEC24C and SEC24D. It recognizes GPI anchors remodeled in the ER by PGAP1 and MPPE1.

In COPI vesicle-mediated retrograde transport, TMED2 inhibits ARFGAP1 activity toward ARF1, preventing premature uncoating and allowing proper cargo selection. It is involved in trafficking of GPCRs such as F2RL1, OPRM1, and P2RY4, contributing to receptor resensitization.

TMED2 facilitates CASR maturation and stabilization in the early secretory pathway, increasing CASR plasma membrane targeting. It may also play a role in maintaining Golgi apparatus organization and biosynthesis of secreted cargo.
