Product Specific Information

The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subpopulations of CD4+ and CD8+ peripheral T lymphocytes, and B cells. CD45RA+ T cell populations are mainly naive/virgin, allowing the use of HI100 mAb as a phenotypic marker to discriminate T cell subsets.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Use in flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometric analysis of normal human peripheral blood cells. Recommended use at 5 µL (0.8 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL of CellBlox Blocking Buffer per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for optimized fluorescence signatures with minimal spillover and spread.

Target Information

CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid cell subtypes. CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It regulates Src-family kinases, crucial for lymphocyte development and antigen signaling.

CD45 isoforms differ by alternative splicing of extracellular domains, influencing membrane localization and signaling behavior. CD45 is involved in:

• Immunoreceptor signaling regulation
• Integrin-mediated signal transduction
• DNA fragmentation during apoptosis

CD45 is expressed on all hematopoietic lineage cells except mature red cells. Dysfunction is associated with conditions such as severe combined immunodeficiency and Hepatitis C.
