Product Specific Information

The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subsets of CD4+ and CD8+ peripheral T lymphocytes and B cells. CD45RA+ T cells are mainly naive, allowing HI100 mAb to serve as a phenotypic marker for T cell subset discrimination.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-titrated and tested on normal human peripheral blood cells
• Recommended Use: 5 µL (0.4 µg) per test, in 100 µL final volume, with 10⁵–10⁸ cells/test

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability testing.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample to antibody cocktails before cell addition.

NovaFluor conjugates utilize Phiton technology, employing nucleic acid dye structures engineered for minimal spectral spillover and spread.

• Excitation: 552 nm
• Emission: 813 nm
• Laser: 561 nm (Yellow)

Strain-dependent observation: NovaFluor Yellow 810 shows non-specific B cell staining in SJL mice, not observed in BALB/c or C57BL/6 strains.

Target Information

CD45RA is an isoform of CD45, a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells. It plays a critical role in lymphocyte development, antigen signaling, and Src-family kinase regulation. Isoform expression varies by cell type and physiological state.

CD45 dysfunction is associated with conditions such as severe combined immunodeficiency and Hepatitis C.
