Target Information
ATG5 (Autophagy Related 5) is an essential protein for autophagy and may play an important role in apoptosis. It is involved in mitochondrial quality control, regulation of innate antiviral response, lymphocyte development, MHC II antigen presentation, and adipocyte differentiation.
Following conjugation to ATG12, the complex participates in autophagosome formation. ATG5 contributes to autophagic cell death via interaction with FADD. The ATG5–ATG12 conjugate forms a cup-shaped isolation membrane that detaches upon autophagosome completion.
APG5 expression occurs downstream of caspase activation during apoptosis. The ATG5–ATG12 conjugate also interacts with innate immune response proteins such as RIG-I and VISA (IPS-1), inhibiting type I interferon production and promoting viral replication.
Dysfunction of ATG5 is associated with spinocerebellar ataxia.
