Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH).
The His tag is commonly expressed at either the N- or C-terminus of recombinant proteins to enable purification via immobilized metal affinity chromatography.
Epitope tagging allows detection of recombinant proteins by fusing a known epitope to them, facilitating identification when specific antibodies are unavailable.
His-tagged proteins are purified using metal chelate chromatography due to the strong and selective affinity of histidine residues for Ni²⁺ ions (e.g., Ni²⁺-NTA resin).
His tag-specific antibodies are used for detection or co-immunoprecipitation of tagged proteins.
