Target Information
CD178 (Fas ligand, FasL)는 type-II membrane protein으로, N-말단은 세포질 내에 위치하고 C-말단은 세포 외부로 확장됩니다. 그 수용체인 FasR은 tumor necrosis factor (TNF) 및 nerve growth factor (NGF) receptor family에 속하며, CD178과 결합 시 apoptosis를 유도합니다.
CD178은 membrane-bound(45 kD) 또는 soluble(26 kD) 형태로 존재할 수 있으며, metalloproteinase에 의해 절단되어 세포 외로 방출될 수 있습니다. Fas와 결합 시 receptor oligomerization을 유도하고, apoptotic cell death를 매개하는 단백질들과 상호작용합니다.
CD178은 활성화된 T-림프구와 NK 세포에서 주로 발현되며, testis 및 eye와 같은 면역 특권 부위에서도 발현됩니다. 또한 thymus, liver, ovary, lung, heart, kidney 등 다양한 조직에서도 보고되었습니다.
CD178을 통한 apoptosis 유도는 항바이러스 면역 반응과 관련이 있으며, FAS/FASLG 시스템은 면역 반응 조절 및 T 세포 매개 세포독성에 관여합니다.
CD178 농도는 동맥경화증 및 염증성 질환, 고혈압 환자에서의 면역 반응 조절과도 관련이 있는 것으로 알려져 있습니다.
Fas/CD178 시스템은 AIDS, hepatitis, cancer 등 다양한 질환에서 중요한 역할을 합니다.
