E2F 전사 인자들은 Rb p110, p107, p130과의 결합을 통해 기능적으로 조절됩니다.
E2F-4는 Rb p110과의 복합체 형성에 의해 조절되며, DP 계열 폴리펩타이드와의 이합체로 DNA에 결합합니다.
E2F-4의 미세이질성은 차등 인산화에 의해 발생하며, 전체 아미노산 길이는 411–416으로 삼염기 반복에 따라 변동됩니다.
