This product is diluted and in a ready-to-use formulation.
Recommended positive control tissues include epithelial cells (pancreas, colon, thyroid, etc.), but are not limited to these.

The antibody is intended for professional laboratory use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissues for manual qualitative immunohistochemistry (IHC) testing. It is used after the primary tumor diagnosis has been made by conventional histopathology.

Cytokeratin 18 (CK18) belongs to the type A (acidic) subfamily of low molecular weight keratins and is found in combination with CK8. Tissues from gastrointestinal, respiratory, and urogenital tracts, as well as endocrine and exocrine tissues and mesothelial cells, are positive for CK18. DC-10 recognizes a variety of simple epithelia including glandular epithelium but not stratified squamous epithelium.

Pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended. IHC staining visualizes antigens via sequential application of primary and secondary antibodies, enzyme complexes, and chromogenic substrates. The visible reaction product is interpreted under a light microscope for differential diagnosis.

A positive tissue control must be run with every staining procedure. This tissue should contain both positive and negative staining components. External positive controls should be fresh specimens processed similarly to patient samples, ideally showing weak positive staining for optimal quality control.

Negative control tissues (internal or external) should demonstrate absence of specific staining to confirm validity. If specific staining occurs in negative controls, patient specimen results must be considered invalid.

Target Information

Cytokeratin 18 (Keratin 18, KRT18, type I cytoskeletal 18) is a type A (acidic) low molecular weight keratin that pairs with cytokeratin 8. It is expressed in non-squamous epithelia and most adenocarcinomas and ductal carcinomas, but not in squamous cell carcinomas. Hepatocellular carcinomas express only CK8 and CK18. Mutations in CK18 are linked to cryptogenic cirrhosis. Two transcript variants encode the same protein. Cytokeratin subtype expression patterns assist in distinguishing epithelial malignancies and are useful in immunohistochemistry, cytopathology, and flow cytometry.
