Target Information
FOXO1 (FKHR, Forkhead Box O1) is a transcription factor belonging to the forkhead homeotic gene family. It can function as a coactivator or corepressor of nuclear receptor activity mediated through the LXXLL motif.
FOXO1 association with PAX3 has been implicated in alveolar rhabdomyosarcoma. It is linked to anti-tumor activity, autophagy, and myogenic growth and differentiation.
FOXO1 is a major target of insulin signaling and regulates metabolic homeostasis in response to oxidative stress. It binds to the insulin response element (IRE) and Daf-16 family binding element (DBE).
FOXO1 regulates redox balance, osteoblast numbers, and bone mass, and modulates endocrine functions related to glucose metabolism. In hepatocytes, it promotes gluconeogenesis by activating genes such as IGFBP1, G6PC, and PPCK1.
FOXO1 acts downstream of CDK1, PKB/AKT1, and SKT4/MST1 as an important regulator of cell death.
