Target Information
FOXO1 (FKHR, Forkhead Box O1) is a transcription factor belonging to the forkhead homeotic gene family. It can function as both a coactivator and a corepressor of nuclear receptor activity through the LXXLL motif. Association with PAX3 is linked to alveolar rhabdomyosarcoma.
FOXO1 plays a role in autophagy, myogenic differentiation, and tumor suppression. It is a key target of insulin signaling, regulating metabolic homeostasis under oxidative stress.
FOXO1 binds to the insulin response element (IRE) and the Daf-16 family binding element (DBE), acting as a regulator of redox balance, osteoblast numbers, and bone mass. It also modulates glucose metabolism by interacting with RUNX2 and PPARGC1A, activating genes such as IGFBP1, G6PC, and PPCK1.
FOXO1 is an important regulator of cell death downstream of CDK1, PKB/AKT1, and MST1.
