Target Information

Peroxiredoxin (Prx) is a growing peroxidase family whose mammalian members are associated with cell proliferation, differentiation, and apoptosis. Many isoforms (about 50 proteins), classified by amino acid sequence homology and active site cysteine residues, exhibit thiol-specific antioxidant activity across all kingdoms.

Mammalian Prx consists of six members grouped into typical 2-Cys, atypical 2-Cys, and 1-Cys Prx types. Except Prx VI (1-Cys subgroup), the other five 2-Cys Prx isotypes possess thioredoxin-dependent peroxidase (TPx) activity utilizing thioredoxin, thioredoxin reductase, and NADPH as a reducing system.

Mammalian Prxs are 20–30 kDa in size and show distinct subcellular localization:

• Prx I, II, and VI: cytosol
• Prx III: mitochondria
• Prx IV: endoplasmic reticulum and secretion
• Prx V: peroxisome, mitochondria, and cytosol
