Target Information

GFI1b (zinc finger protein GFI-1b) is an essential proto-oncogenic transcriptional regulator necessary for development and differentiation of erythroid and megakaryocytic lineages. It is a component of the RCOR-GFI-KDM1A-HDAC complex that suppresses genes involved in multilineage blood cell development through histone deacetylase (HDAC) recruitment, thereby controlling hematopoietic differentiation.
GFI1b functions as a transcriptional repressor or activator depending on promoter and cell type context. It represses SOCS1 and SOCS3 promoter activity, regulating cytokine signaling pathways, and cooperates with GATA1 to repress target gene transcription such as BCL2L1. Silencing of GFI1B in leukemic cell lines markedly increases apoptosis rate.
Mutations affecting the gene are associated with Bleeding disorder, platelet-type 17 (BDPLT17).
