Target Information

Ankyrins are a family of proteins that link integral membrane proteins to the underlying spectrin–actin cytoskeleton, playing key roles in cell motility, activation, proliferation, contact, and maintenance of specialized membrane domains. They are composed of three structural domains:

• An amino-terminal domain with multiple ankyrin repeats
• A central spectrin-binding domain
• A carboxy-terminal regulatory domain, which is variable and subject to alternative splicing

Ankyrin 1, the prototype of this family, was first identified in erythrocytes and later found in brain and muscle tissues. Mutations in erythrocytic ankyrin 1 are associated with approximately half of all hereditary spherocytosis cases. Various isoforms and truncated muscle-specific variants have been identified due to alternative promoter usage and splicing events.
