Target Information
Smoothened drosophila homolog (SMOH) is a serpentine protein belonging to the G-protein coupled receptor Fz/Smo family with 787 amino acids. It contains seven transmembrane domains, four glycosylation sites, and a putative extracellular amino terminus. Smoothened, activated by β-Arrestin 2 and GRK2, acts as a receptor for Shh (Sonic hedgehog gene product) and is a positive regulator of the hedgehog signaling pathway along with the patched protein (PTCH). It regulates vertebrate developmental processes, cellular proliferation, and differentiation by activating transcription factor Gli. Mutations in Smoothened are known to cause sporadic basal cell carcinoma of the skin. Expression is widespread in heart and brain.
