Voltage-dependent calcium channels mediate the entry of calcium ions into excitable cells and are involved in various calcium-dependent processes such as muscle contraction, hormone or neurotransmitter release, and gene expression.
These channels are multisubunit complexes composed of alpha-1, beta, alpha-2/delta, and gamma subunits. The alpha-1 subunit forms the pore and determines channel activity, while auxiliary subunits regulate this function.
The calcium channel types differ mainly by expression of alpha-1 isoforms (A, B, C, D, E, S). The CACNA1A gene encodes the alpha-1A subunit, predominantly expressed in neuronal tissue. Mutations are associated with familial hemiplegic migraine and episodic ataxia type 2.
Polymorphic variation due to n-repeats occurs; some transcript variants include a polyglutamine tract, and expansion of these repeats (from normal 4–16 to 21–28) is linked to spinocerebellar ataxia type 6.
