Product Specific Information
MA1-2013 detects beta adrenergic receptor kinase 1 (GRK2) in human, mouse, and rat samples.
It has been successfully applied in Western blot, immunocytochemistry, immunofluorescence, immunoprecipitation, and ELISA assays.
In Western blot, it detects a ~30 kDa protein representing beta adrenergic receptor kinase 1 in human HeLa cells.
In immunocytochemistry, MA1-2013 recognizes the target in HeLa, HEK-293, and 3T3 cells, showing predominantly cytoplasmic membrane-associated staining and ruffle localization in immunofluorescence.
The immunogen used is recombinant beta adrenergic receptor kinase 1.
Target Information
ADRBK1 (GRK2) is a serine/threonine kinase that phosphorylates G protein-coupled receptors (GPCRs).
GRK2 (83 kDa) is one of six GRK family members expressed ubiquitously in mammals.
Upon ligand binding, GPCRs are phosphorylated by GRKs, leading to internalization mediated by beta-arrestin-1 and activation of the Ras–Raf–ERK1&2 signaling pathway.
GRK2 activity is regulated by phosphorylation through PKC, Src, and ERK1&2, as well as interactions with various proteins.
ERK phosphorylates GRK2 on serine 670, inactivating it via a negative feedback mechanism.
