Product Specific Information
The MBSA43 monoclonal antibody reacts with human TIGIT (T cell immunoreceptor with Ig and ITIM domains), also known as VSTM3 or WUCAM. TIGIT is a 26 kDa protein belonging to the poliovirus receptor (PVR) family. It is expressed on NK cells, regulatory T cells, follicular T helper cells, memory CD4+ and CD8+ T cells, but not on B cells or naïve CD4+ T cells. Expression can be upregulated upon activation.
MBSA43 is reported to cross-react with Rhesus macaque.

- Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells
- Recommended Usage: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)
- Light Sensitivity: Protect from light; tandem dye is sensitive to photo-induced oxidation
- Fixation Compatibility: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) with minimal fluorescence loss
- Excitation: 488–561 nm
- Emission: 775 nm
- Laser Compatibility: Blue, Green, Yellow-Green lasers
- Filtration: 0.2 µm post-manufacturing filtered
Target Information
TIGIT binds with high affinity to the poliovirus receptor (PVR), leading to increased IL10 secretion and decreased IL12B secretion, thereby suppressing T cell activation and promoting the generation of mature immunoregulatory dendritic cells.
