Target Information
Smad4는 인간 염색체 18에 위치한 Smad4 유전자에 의해 암호화된 단백질로, Smad family member 4, Sma- and Mad-related protein 4 또는 Mothers against decapentaplegic (MAD) homolog 4로도 알려져 있습니다.
Smad4는 DNA 결합과 단백질 상호작용에 관여하는 N-말단 MH1 (MAD homology 1)과 C-말단 MH2 (MAD homology 2) 도메인을 포함합니다.
TGF-beta 및 BMP 등의 리간드가 수용체에 결합하면 Smad 1, 2, 3, 5, 8 (R-Smad)이 인산화되어 Smad4(co-Smad)와 삼합체를 형성하고 핵으로 이동합니다.
Smad 6, 7과 같은 억제성 Smad(I-Smad)는 활성화된 R-Smad와 상호작용하여 신호 전달을 억제합니다.
Smad4는 Cyclin D1 억제 및 Collagen 발현 증가를 통해 세포 증식을 억제하는 종양 억제 단백질로 작용합니다.
탈인산화는 Smad의 핵 내외 이동 및 동적 균형을 조절합니다.
