Target Information

FOXK1 is a transcriptional regulator involved in glucose metabolism, aerobic glycolysis, muscle cell differentiation, and autophagy. It recognizes and binds the forkhead DNA sequence motif (5'-GTAAACA-3') and can act as both activator and repressor depending on context.
Together with FOXK2, it regulates metabolic reprogramming toward aerobic glycolysis, promoting expression of glycolytic enzymes such as HK2, PFK, PKLR, and LDH, while suppressing mitochondrial pyruvate oxidation via PDK1 and PDK4.
It plays roles in gluconeogenesis during fasting, mTORC1-mediated metabolic regulation, and autophagy inhibition in skeletal muscle. FOXK1 also regulates myogenic progenitor cells, represses FOXO4 activity, and modulates Wnt/β-catenin signaling.
Additionally, it contributes to antiviral responses by binding ISRE to promote antiviral gene expression. [UniProt]
