Target Information
FOXK1 is a transcriptional regulator involved in glucose metabolism, aerobic glycolysis, muscle differentiation, and autophagy.
It binds the forkhead DNA motif (5'-GTAAACA-3') and can function as both activator and repressor depending on context.
Together with FOXK2, it regulates metabolic reprogramming toward aerobic glycolysis by promoting expression of glycolytic enzymes (HK2, PFK, PKLR, LDH) and inhibiting pyruvate oxidation via upregulation of PDK1 and PDK4.
FOXK1 participates in mTORC1-mediated metabolic reprogramming, autophagy regulation in skeletal muscle, and muscle repair processes.
It also enhances Wnt/β-catenin signaling and may promote antiviral gene expression by binding ISRE elements.
