Description:
The 22URTI monoclonal antibody reacts with human interleukin (IL)-22, a 20 kDa member of the IL-10 cytokine family secreted primarily by Th17 and NK cells. Other T cells can also produce IL-22. In Th17 cultures, IL-22 expression is induced more strongly by IL-23 than by IL-6 or TGF-β, suggesting secretion by mature Th17 cells in vivo. The IL-22 receptor is a heterodimer of IL-10R2 and IL-22R1.

Applications Reported:
This antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for intracellular staining and flow cytometric analysis of human peripheral CD4 cells cultured in Th17 polarizing conditions. Use at 5 µL (0.06 µg) per test. A test is defined as the amount of antibody that stains a cell sample in 100 µL final volume (typically 10⁵–10⁸ cells/test).

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness or FRET efficiency.

Target Information

IL-22, also known as IL-10-related T-cell derived inducible factor, is an alpha-helical cytokine and member of the IFN-IL-10 family (including IL-19, IL-20, IL-24, IL-26, IL-28, IL-29, and type I/II interferons). Produced mainly by activated T cells and NK cells, IL-22 is encoded on chromosome 12q and structurally related to IL-10.

IL-22 binds to a heterodimeric receptor complex (IL-22R1 and IL-10R2) to activate the JAK-STAT signaling pathway. IL-22BP, a soluble receptor, antagonizes IL-22 binding. IL-22 primarily targets nonimmune cells (skin, digestive and respiratory tissues, hepatocytes, keratinocytes) and plays roles in innate immunity, wound healing, and host defense against extracellular pathogens. It is upregulated during chronic inflammation and mediates IL-23-induced dermal inflammation via STAT3 activation.
