Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) act as adaptor proteins linking TNF receptor superfamily members to intracellular signaling pathways, regulating cell death and stress responses. TRAF proteins share a conserved region that mediates interactions with receptors and other TRAF proteins, leading to activation of diverse signaling cascades.
TRAF2 and TRAF3 directly bind to CD40, a receptor involved in B cell immunity, and are critical for NF-κB activation in mouse B lymphocytes. TRAF2 also functions with TRAF6 in CD40 signaling in non-hematopoietic cells and interacts with the lymphotoxin-beta receptor (LTβR) along with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
