Target Information

SMAD2 (MADH2, MAD2) regulates multiple cellular processes such as cell proliferation, apoptosis, and differentiation.
It interacts with TGF-beta receptors via SMAD anchor for receptor activation, leading to nuclear translocation—a central event in TGF-beta signaling.
Phosphorylation of threonine 8 by ERK1 enhances SMAD2 transcriptional activity, while calmodulin negatively regulates it.
Upon TGF-beta stimulation, SMAD2 is phosphorylated by TGF-beta receptors, dissociates from SARA, and associates with SMAD4 for nuclear translocation and transcriptional regulation.
SMAD2 can also be phosphorylated by activin type 1 receptor kinase to mediate activin signaling.
Alternatively spliced transcript variants encoding SMAD2 have been reported.
