Target Information

This gene encodes a member of the tumor necrosis factor (TNF) cytokine family which acts as a ligand for osteoprotegerin and plays a key role in osteoclast differentiation and activation. It also functions as a dendritic cell survival factor and regulates T cell-dependent immune responses. T cell activation induces expression of this gene, promoting osteoclastogenesis and bone loss.

The protein activates antiapoptotic kinase AKT/PKB through a signaling complex involving SRC kinase and TRAF6, suggesting a role in apoptosis regulation. Gene disruption in mice results in severe osteopetrosis, absence of osteoclasts, and defects in early T and B lymphocyte differentiation. Deficient mice also fail to form lobulo-alveolar mammary structures during pregnancy. Two alternatively spliced transcript variants have been identified.
