Target Information
Follistatin-like protein 1 (FSTL1) is a widely expressed extracellular glycoprotein belonging to the osteonectin (BM-40/SPARC) family. It contains both a follistatin-like domain and an extracellular calcium-binding domain. Initially identified as a TGF-beta-inducible protein in a mouse osteoblast cell line, FSTL1 participates in multiple cellular functions such as proliferation, differentiation, apoptosis, and migration. It is involved in biological processes including embryonic development, inflammatory response, angiogenesis, tumorigenesis, and immune disease pathogenesis.
FSTL1 expression is highly conserved across mammals and is widely present in human tissues. It can be upregulated by innate immune mediators such as TLR4 agonists and IL-1β via NFκB pathways, leading to increased secretion of pro-inflammatory cytokines (TNF-α, IL-1β, IL-6, IL-8). While mesenchymal cells produce FSTL1, hematopoietic cells typically do not under normal conditions. At sites of inflammation, macrophages and monocytes can uptake FSTL1, inducing caspase-1 activation and IL-1β maturation. Overexpression of FSTL1 contributes to immune diseases such as rheumatoid arthritis and osteoarthritis, whereas reduced serum levels are associated with ovarian and endometrial cancers, affecting cell proliferation and migration.
