Target Information

T-box 3 (TBX3) is a member of a conserved gene family sharing a common DNA-binding domain, the T-box. These genes encode transcription factors that regulate developmental processes.
Tbx3 functions as a transcriptional repressor binding to canonical Brachyury binding sites and plays a role in anterior/posterior axis formation of the tetrapod forelimb. It is essential for normal mammary development and is implicated in tumorigenesis.
Tbx3 expression increases during osteoblast differentiation and may influence osteoblast cell numbers. Mutations in T-box 3 cause ulnar-mammary syndrome (MIM 181450), affecting limb, gland, tooth, hair, and genital development.
Alternative splicing generates three transcript variants encoding different isoforms. Overexpression of Tbx3 has been observed in certain human breast cancer cell lines and can induce immortalization in mouse embryo fibroblasts. Truncated forms of Tbx3 are elevated in plasma samples.
